日本語
実際の肺NTM患者から気道上皮細胞を採取して線毛運動などの解析を行うことで、宿主側の細胞機能という側面から感染機序や患者背景の解明を行う。また、これら臨床検体の気道上皮細胞機能解析の問題点として、細胞機能(線毛運動)の異常が「NTM感染の原因なのか、あるいはNTM感染によって引き起こされた結果なのか、という所謂「鶏と卵」の関係」の疑問があった。
そこで、肺NTM患者の気道上皮細胞を末梢血からiPS細胞の技術を用いてin vitroで再現することで、宿主側の感染や他喫煙曝露等の気道上皮細胞機能に影響を与える後天的な因子を除いた系で機能評価を実施して、直接的に患者から採取された気道上皮細胞と比較することで、より本質的なメカニズムの解明を目指す。

Airway epithelial cells are collected directly from patients with NTM-LD by bronchoscopy and analysed for lineage movement, etc., to elucidate the infection mechanism and patient background from the perspective of cell function on the host side. One problem with the analysis of airway epithelial cell function in clinical samples was the question of whether abnormalities in cell function (fibrillary motility) were the cause of NTM infection or the result of NTM infection, in a so-called 'chicken and egg' relationship.
Therefore, by reproducing airway epithelial cells from patients with NTM-LD in vitro using iPS cell technology from peripheral blood, functional evaluation was carried out in a system excluding acquired factors affecting airway epithelial cell function, such as infection on the host side or exposure to other smokes, and by comparing them with airway epithelial cells directly from the patient, Aiming to elucidate more essential mechanisms.
